Time Flies! Partying in Laguna!

Time really does fly! I keep forgetting to write here at What is up with that?

I guess I can do a quick summary of things.

I wnet to a neighbors pary last night.

They have a killer house here in Laguna Beach and it made for throwing parties! I live in a nice place - but their house is designed for the flow of partying activity - which is very cool!

They have a multilevel back yard and you walk down to the back which is under the house down some steps. It has a veranda with stoves and stuff and is perfect to have the caterers serving up food and they had a nice buffer.

They had a "one man band" which is not as good as a real band - but he did play the corqwd pleasers most drunk folks like to hear - so i guess it was ok. I'd prefer some real musicians jamming it up at least a little.

They also had a "couple" teach "Tango lessons". That was weird! What is up with that?

Maybe not my slice of pie - but the drunk partyers seems to like it!

Well - that was my day yesterday! At least now I might remember more of it!

It's warm here but not too hot!

Wow - time really flies! I just noticed it has been a long time since I last posted here. I feel bad. I am embarrassed. But I am here now so let's make up for all that lost time...

It's May 2010 and I am living in Laguna Becah, California.

I'm really digging it here. The waether is simply awesome! Almost every day is right around 70 degrees. Of course we get some hot days in the upper 80's now and again, but for the most part we are right around 70.

Some people may look at 70 and say "that's too cold", but hey - it almost NEVER feels cold here. That's right - even in the winter when everyone else really is COLD _ I mean REALLY FEELING COLD - we're still right around 70 more than likely.

Sure - every once in a while the nights will drop a few degrees - but since we are on the Pacific Ocean - it never gets REALLY COLD.

So - as far as climate - I thk this is the best place on earth!

I know - some people think the best climate is a HOT and SWEATY one, but I will say that after having lived in the San Fernando Valley - in Sherman Oaks to be exact - it was so hot so often it was NOT COMFORTABLE! It was uncomfortable most of the time. And sure - it did get HOT there a lot - but it also got VERY COLD there a lot. It was inland so both HIGHS and LOWS were more extreme.

I'm not saying it was BAD. I'm not - really. While I lived therre (almost 10 years) I did no t know any better. I accepted the many 90 and 100 degree days and the cold winter nights as being normal. But man - when I discoverd what it is like to lkive on the Southern California coast - my eyes opened to what comfortable really is all about!

Well - enough about the climate. I do dig it here a lot! It's a great place to live here in Laguna Beach. Some say it magical. I agree!

Planning a Fiesta BBQ Party

Fiesta BBQ Party Planning Tips

BBQs are popular for events such as the 4th of July, birthday parties, receptions, and going away fiesta parties. No matter why you have decided to plan a barbeque fiesta party, the memories that will come from it are sure to stick with your guests for many years to come, provided that you plan ahead. Alternatively, a new popular type of barbeque is a BBQ clambake. Clambakes are typically held in a pit on the beach, but if a beach is not available, you may want to consider holding your clambake right from your grill.

1) First, no barbeque would be complete without guests. Plan ahead of time, checking with your potential guest for fiestascheduling conflicts and issues. Finding a date and time that is suitable for everyone should not be to far as long as you everyone enough advanced notice about the party.

Fiesta planning is critical!

Remember that not everyone will be able to come to your BBQ fiesta, no matter how much notice you give them for the party, so do not get discouraged if you do not get as many RSVPs as your originally hoped for.

2) Be sure to check the weather for that type of year. If you live in a climate that cannot be predicted, then you may want to have a backup plan just incase unusual weather comes. You cannot predict things like rain and thunderstorms ahead of time, but you can make sure that you hold you barbeque during the time of year that will be pleasant to your guests. A BBQ in cold weather may hold more negative memories of trying to keep warm rather then good memories of family and friends.

3) You will need to purchase the items for your clambake. The traditional items you will need is Clams still in the shell, seawater(if available), corn still in the husk, and potatoes.

Side dishes can also be bought, but are not necessary since the potatoes and corn can act as such. If you feel that you need other side dishes, the simplest approach to side dishes is chips and dip. Chips and dip do not require any cooking on your part, and will still be loved by the majority of your guests.

4) Depending on the theme of your BBQ party, you may or may not want to purchase decorations. Festive decorations can be found at any local party or general store. You may also consider purchasing decorations that can be stored and reused at a different time, such as reusable tablecloths and Tiki torches. Give your clambake a beach theme by adding tropical music and you may even consider adding some sand.

5) The day before you BBQ party, you will want to make sure to thaw, prepare, and marinate the meat you will be cooking. Store the meat stored away, soaking up the marinate until it is time for it to go on the grill. Prepare any side dishes that you can ahead of time.

6) The day of the BBQ party, get up early and start preparing for it. You do not want your guest arriving while you are still cooking or hanging decorations. It should only take you a few hours at most to prepare the area in which you will be having the party, but the reactions from your guest will probably seem as if you have been planning this for weeks.

Place your clambake on the grill, layering your already cooked potatoes, corn still in the husk, and your clams on the grill. Then grill these until the corn and clams are complete.

Depending on the time in which your guests arrive, you may want to include you guests in the actual cooking of the BBQ. Enjoying a few drinks around the pit or stove will not only give you time to gather and converse, but will also make your guests hungry, as they smell the wonderful aroma coming from the steaming or boiling vegetables, steaks and clams.

Genetic Screening As Preimplantation Genetic Diagnosis - Increased IVF Success

An inability to establish or maintain pregnancy is most often attributed to lethal genetic defects of the embryo. For infertile couples and carriers of genetic disease, the option of in vitro fertilization (IVF) can often offer the only viable alternative. Over the last 15 years, advances in molecular genetics has allowed for embryos to be genetically screened prior to implantation. Employing various genetic techniques such as Polymerase Chain Reaction (PCR), Fluorescence in situ Hybridization (FISH) and Comparative Genomic Hybridization (CGH), this process - known as preimplantation genetic diagnosis (PGD) - is now used to identify embryos with a range of genetic abnormalities such as chromosomal aneuploidy, single gene defects and X-lined disorders. This article will discuss some advantages and disadvantages of using this technology in IVF assisted reproduction.

The use of PGD ensures that only healthy embryos are implanted in the womb. This presents a number of obvious advantages. Firstly, it substantially increases the chance of successful pregnancy by eliminating genetic factors which cause subsequent miscarriage. It also avoids the trauma of multiple terminations following unfavorable prenatal diagnoses - a reality that is all too common for couples with genetic disease. Ultimately, as well as diminishing emotional and financial stress of repeated IVF cycles, PGD ultimately prevents the birth of individuals with genetic disease. These individuals would otherwise face a myriad of medical and social problems which would both reduce their quality of life and significantly reduce their life expectancy.

The application of PGD to IVF presents other favorable opportunities. Albeit controversial, embryos can be screened for histocompatibility to identify a match for a sick sibling. This can often be the best opportunity - or indeed the only opportunity - to save the life of the sibling with a terminal illness, although this may often present ethical and moral considerations. Furthermore, embryos rejected for implantation can, with appropriate consent, be donated for medical research. This would help to alleviate the current lack of stem cells which are needed to develop future treatments for inherited disease.

PGD has clear advantages but the process is not without limitations. Many studies indicate that it has little or no effect for mature (age 40+) women trying to conceive. Also, since PCR and FISH usually analyze single cells, there is a possibility of false positive and false negative results - errors which are usually due to multiple cell lines in the early embryo through disjunction at mitosis. Consequently, healthy embryos are occasionally discarded as aneuploidic before they have the opportunity to 'self-correct', a phenomenon known to occur in these so called 'mosaic' embryos.

Other limitations of PGD include failure to detect rare types of polygenic disease, such as those of cystic fibrosis which has over 1000 known mutations. Even the basic cell extraction is in itself technically challenging. The embryo must be held in position by a holding pipette, while the zona pellicuda is breached using a micro-needle before a single cell can removed by aspiration. This is a delicate process which poses a risk of irreversible damage to the developing embryo and requires considerable training. Coupled with the fact that the process is expensive and therefore not available to everyone, it is clear that there are also disadvantages.

PGD may give couples with genetic disease the opportunity to produce healthy offspring but the technologies have potential applications which society may deem ethically questionable. For example, is it morally or ethically sound to 'edit' which children should be born into the world? Will genetically affected embryos always be used in the manner for which consent is given? Should PGD be used for non-medical reasons such as selection for desired traits or gender? These are questions that society must surely address as this technology develops.
